Target Information
The accumulation of unfolded proteins in the endoplasmic reticulum (ER) triggers the ER stress response. This response includes:
- Inhibition of translation to prevent further accumulation of unfolded proteins
- Increased expression of proteins involved in folding (unfolded protein response, UPR)
- Degradation of misfolded proteins via the ER-associated protein degradation (ERAD) system
HERPUD1 may play a role in both UPR and ERAD. Its expression is induced by ER stress and contains an ER stress response element in its promoter region. The encoded protein has an N-terminal ubiquitin-like domain that may interact with the ERAD system. HERPUD1 interacts with presenilin proteins and can increase amyloid-beta levels upon overexpression. Alternative splicing produces multiple transcript variants with differing isoforms.
