Target Information
IFI16 (gamma-interferon-inducible protein 16) binds double-stranded DNA, preferentially to supercoiled and cruciform DNA structures. It is involved in transcriptional regulation and acts as a transcriptional repressor. IFI16 may regulate hematopoietic differentiation and cell proliferation through modulation of p53/TP53 and retinoblastoma protein functions. Additionally, IFI16 participates in innate immune responses by recognizing viral dsDNA in the cytosol and nucleus, recruiting TMEM173/STING, and inducing IFN-beta production. It is essential for activating the IFR3 signaling cascade during HSV-1 infection and contributes to heterochromatin assembly on viral DNA, inhibiting viral gene expression and replication. IFI16 also plays a role in MTA1-mediated epigenetic regulation of ESR1 expression in breast cancer.
