Target Information

14-3-3 sigma (stratifin, SFN, epithelial cell marker protein 1) is a member of a highly conserved family of 14-3-3 proteins found in all eukaryotic organisms. There are seven known mammalian isoforms (epsilon, beta, zeta, gamma, theta, tau, sigma), which play critical roles in biological activities by binding and modifying the localization or stability of key molecules in various signaling cascades.
14-3-3 sigma was first identified as a human mammary epithelium marker and later recognized for its role in cell cycle checkpoint regulation. It may serve as a prognostic marker for pancreatic cancer survival. This isoform is uniquely induced by the tumor suppressor protein p53 in response to DNA damage and acts as a p53-regulated inhibitor of G2/M progression, functioning as a tumor suppressor gene that can be silenced by methylation in epithelial tumor cells. Two isoforms of human 14-3-3 sigma are produced by alternative splicing.
