Target Information

14-3-3 sigma (stratifin, SFN, epithelial cell marker protein 1) is a member of a highly conserved family of 14-3-3 proteins present in all eukaryotic organisms. There are seven known mammalian 14-3-3 isoforms (epsilon, beta, zeta, gamma, theta, tau, sigma) that play important roles in various biological activities by binding to and altering the localization or stability of key molecules in signaling cascades.

14-3-3 sigma was originally characterized as a human mammary epithelium marker and later identified as a molecule involved in cell cycle checkpoint regulation. It may serve as a prognostic marker for pancreatic cancer treatment outcomes. It is the only isoform induced by tumor suppressor protein p53 in response to DNA damage and acts as a p53-regulated inhibitor of G2/M progression, functioning as a tumor suppressor gene that can be inactivated by methylation of its CpG islands in epithelial tumor cells. Two isoforms of human 14-3-3 sigma are produced by alternative splicing.
