Product Specific Information

Description:
The RM4-5 monoclonal antibody recognizes the mouse CD4 molecule, a 55 kDa cell surface receptor expressed on most thymocytes, subsets of mature T cells, and dendritic cells. CD4 binds to MHC class II molecules on antigen-presenting cells and is crucial for T cell development and function. RM4-5 binding is blocked by GK1.5.

Applications Reported:

• Reported for use in flow cytometric analysis.

Internal Testing Notes:

• Alexa Fluor 561 may non-specifically stain B cells in Swiss Webster and SJL mice.
• No non-specific staining observed in BALB/c or C57BL/6 mice.
• Other strains not tested.

Applications Tested:

• Tested by flow cytometric analysis of mouse splenocytes.
• Recommended ≤0.125 µg per test (100 µL final volume).
• Optimal cell number: 10⁵–10⁸ cells/test.
• Titrate for best performance.

Target Information

CD4 antigen is involved in recognition of MHC class II molecules and acts as a co-receptor for HIV. It is mainly expressed on T helper lymphocytes, and also found on monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and some non-lymphoid tissues such as brain and gut. CD4 functions in T-cell activation via association with the T-cell receptor complex and Lck kinase, and may mediate neuronal damage in immune-related CNS diseases.
