Target Information
The mediator complex is a multi-protein transcriptional co-activator expressed ubiquitously in eukaryotes and required for induction of RNA polymerase II transcription by DNA binding transcription factors.
MED28, also known as Magicin, is one of the proteins in this complex and is expressed in many cell lines and tissues. Down-regulation of MED28 in NIH3T3 cells induces genes associated with smooth muscle cell differentiation, while over-expression represses SMC gene expression.
MED28 can form a ternary complex with Grb2 and Merlin (the neurofibromatosis 2 tumor suppressor protein), suggesting a role in tumor suppressive activity. It has also been identified as an HIV dependency factor (HDF), indicating potential as a drug target in HIV research.
At least two isoforms of MED28 are known to exist.
