Target Information
The mediator complex is a multi-protein transcriptional co-activator expressed ubiquitously in eukaryotes and required for induction of RNA polymerase II transcription.
MED28 (also known as Magicin) is one of the proteins in this complex and is expressed in many cell lines and tissues. Down-regulation of MED28 in NIH3T3 cells induces genes associated with smooth muscle cell differentiation, while overexpression represses these genes.
MED28 forms a ternary complex with Grb2 and Merlin (NF2 tumor suppressor protein), suggesting a role in tumor suppression. It has also been identified as an HIV dependency factor (HDF), indicating potential as a drug target in HIV treatment.
At least two isoforms of MED28 are known to exist.
