Product Specific Information
This antibody is predicted to react with human and some non-human primates based on 100% sequence homology.
It is specific for the ~53 kDa p53 protein phosphorylated at Ser392 in Western blots of NIH 3T3 cell lysates.
Immunolabeling is blocked by the phosphopeptide used as the antigen but not by the corresponding dephosphopeptide.
Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress.
It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation.
p53 activates transcription of genes such as p21waf1, leading to G1 or G2/M arrest.
The protein includes an N-terminal transactivation domain, a DNA-binding domain, an oligomerisation domain, and a C-terminal regulatory domain.
Phosphorylation at Ser392 is associated with human tumor formation.
p53 also relates to aging, oxidative stress, and cognitive function deficits linked to LTP (Long Term Potentiation).
In normal cells, p53 is expressed at low levels, while in transformed cells, it is highly expressed and contributes to malignancy.
Mutations in TP53 lead to loss of tumor suppressor activity and are found in many cancers, including germline mutations in Li-Fraumeni syndrome families.
