Product Specific Information

The 104D2 monoclonal antibody reacts with human CD117 (c-Kit), also known as Steel factor receptor and stem cell factor receptor. This 145 kDa tyrosine kinase receptor is expressed by hematopoietic progenitor cell subsets and mast cells. Interaction of c-Kit and Steel factor promotes proliferation and differentiation of hematopoietic progenitor cells and mast cell differentiation. CD117 is also expressed by melanocytes and plays a role in their activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test in a final volume of 100 µL. Cell number: 10⁵–10⁸ cells/test.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Recommended filter: 710/50 bandpass; alternative: 695/40 bandpass.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-822-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and compensation.

Target Information

KIT (c-KIT) is a proto-oncogene and type 3 transmembrane receptor for MGF (mast cell growth factor, stem cell factor). KIT regulates growth and activation of hematopoietic and non-hematopoietic cells. Mutations in KIT are associated with gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism. Activation of c-Kit by SCF leads to receptor dimerization, autophosphorylation, and downstream signaling through Ras/ERK, PI3K, PLC-γ, and JAK/STAT pathways. KIT receptor activation is essential for hematopoiesis, stem cell maintenance, and gametogenesis.
