Product Specific Information

The AFS98 monoclonal antibody reacts with mouse CD115 (M-CSF receptor, c-fms). CD115 is expressed by monocytes, macrophages, osteoclasts, and some epithelial cells. It is a 150 kDa product of the c-fms gene and belongs to the immunoglobulin family.
CSF-1 signaling through CSF-1R regulates proliferation and differentiation of monocytic lineage cells.

• Applications Tested: Tested on mouse thioglycolate-induced peritoneal exudate cells at ≤0.06 µg/test
  • 1 test = antibody amount staining a 100 µL cell sample
  • Cell number range: 10⁵–10⁸ cells/test
  • Optimize antibody titration for best performance

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by the blue laser (488 nm).
Recommended filter: 710/50 bandpass (695/40 acceptable). Ensure instrument compatibility.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222-49)
• 1-step Fix/Lyse Solution (Product #00-5333-54)
  Clone-specific performance after fixation should be verified empirically.

CSF1R (FMS) is a tyrosine kinase receptor involved in macrophage production, differentiation, and function.

• Activated by ligand binding through oligomerization and transphosphorylation
• Member of the CSF1/PDGF receptor family
• Mutations linked to myeloid malignancy predisposition
• Regulates survival, proliferation, and differentiation of hematopoietic precursor cells, especially mononuclear phagocytes
• Promotes release of proinflammatory chemokines in response to IL34 and CSF1
• Plays a key role in innate immunity and inflammatory processes
