The monoclonal antibody 51.1 reacts with human CD1d, a member of the CD1 family similar to non-polymorphic MHC Class I-like molecules. CD1d is a conserved transmembrane receptor of the Immunoglobulin Superfamily and can associate with beta-microglobulin or exist independently, suggesting different mechanisms for glycolipid antigen presentation to CD1d-reactive NKT cells.
CD1d expression is found on peripheral B cells, resting monocytes, cortical thymocytes, and intestinal epithelial cells (IEC) with polarized expression. It is also found at low levels in hepatocytes and is highly expressed in HCV-infected livers compared to normal controls.
The 51.1 monoclonal antibody has functional activity, blocking interactions between CD1d-transfected cells and NKT cells.

Target Information
CD1d is a heavy chain associated with Beta 2 microglobulin on cortical thymocytes. Beta 2 microglobulin-independent expression has been shown on human intestinal epithelial cells. CD1d is the sole group 2 member of the CD1 family of MHC-like glycoproteins. The CD1d gene encodes a transmembrane glycoprotein forming heterodimers with beta-2-microglobulin and mediating lipid/glycolipid antigen presentation to T cells.
The human genome contains five CD1 family genes clustered on chromosome 1. These members differ in localization and lipid ligand specificity. CD1d localizes to late endosomes and lysosomes via a tyrosine-based motif in its cytoplasmic tail.
