Description:
The OX85 monoclonal antibody reacts with rat CD62L (L-Selectin), a 76-kDa member of the selectin family expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. CD62L binds glycosylated, fucosylated, sulfated, and sialylated glycoproteins such as CD34, GlyCAM-1, and MadCAM-1, mediating lymphocyte rolling on activated endothelium and homing to high endothelial venules of peripheral lymphoid tissues.

Applications Reported:
This OX85 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of rat splenocytes. Use ≤0.25 µg per test (100 µL final volume). Optimal cell number (10⁵–10⁸ cells/test) should be determined empirically. Antibody titration is recommended for optimal performance.

Fluorophore Information:
eFluor® 660 replaces Alexa Fluor® 647. It emits at 659 nm and is excited with the red laser (633 nm). Ensure the instrument can detect this fluorochrome.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein of the selectin family of adhesion receptors. It includes an N-terminal C-type lectin domain, EGF-like domain, two SCR sequences, a short spacer, transmembrane, and cytoplasmic regions. CD62L is constitutively expressed on most leukocytes including lymphocytes, monocytes, and polymorphonuclear cells, except for a subset of memory T cells.
