Product Specific Information

Description:
The 2C9.G3 (HMb3-1) monoclonal antibody reacts with mouse and rat CD61 (integrin beta3). CD61 is expressed by activated T cells, granulocytes, and platelets. It associates with alphaV (CD51) to form the Vitronectin Receptor and with alphaIIb (CD41) to form gpIIb/IIIa. These complexes mediate adhesion to extracellular matrix components including fibrinogen, fibronectin, vitronectin, thrombospondin, and von Willebrand factor.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes and bone marrow cells at ≤ 0.2 µg/test (100 µL final volume). Optimal titration is recommended.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on engineered nucleic acid dye structures for optimized fluorescence signatures with reduced spillover and spread.

A 105 kDa glycoprotein expressed on megakaryocytes, platelets, and their precursors.
Functions as a receptor for fibrinogen, fibronectin, von Willebrand factor, and vitronectin.
Forms complexes with alphaIIb (CD41) or alphaV (CD51), participating in cell adhesion and signaling.
Associated disorders include Glanzmann Thrombasthenia and Platelet type-16 Bleeding Disorder.
