Target Information
BID is a death agonist that heterodimerizes with either agonist BAX or antagonist BCL2. It belongs to the BCL-2 family of cell death regulators and mediates mitochondrial damage induced by caspase-8 (CASP8). CASP8 cleaves BID, and the COOH-terminal fragment translocates to mitochondria to trigger cytochrome c release. BID acts as a pro-apoptotic molecule linking caspase-8 activation to mitochondrial apoptosis pathways. In living cells, BID typically exists in an inactive cytosolic form and becomes cleaved and activated by caspase-8 in response to TNF-alpha or Fas ligand stimulation. The cleaved 15 kDa fragment (p15) translocates to mitochondria and induces cytochrome c release, leading to apoptosis.
