The 1H8PWSR monoclonal antibody reacts with mouse interleukin (IL)-22, a 20 kDa cytokine in the IL-10 family secreted primarily by Th17 and NK cells. IL-22 expression is induced by IL-23 and acts through the IL-10R2/IL-22R1 receptor complex to stimulate antimicrobial peptide expression such as β-defensin-2 and -3. IL-22 plays a protective role against bacterial infections in lungs and gut.

• Applications Reported: Intracellular staining followed by flow cytometric analysis.
• Optimized for: eBioscience IC Fixation and Permeabilization Kit (Product #88-8823-88).
• Applications Tested: Th17-polarized mouse splenocytes restimulated with PMA, Ionomycin, and Brefeldin A for 5 hours.
  • ≤ 0.125 µg/test in 100 µL final volume.
  • Cell number: 10⁵–10⁸ cells/test (empirically determined).
  • Titrate for optimal performance.

Target Information

IL-22 (also known as IL-10-related T-cell derived inducible factor) is an α-helical cytokine in the IFN-IL-10 family. It is produced mainly by activated T cells and NK cells and acts via a receptor complex of IL-22R1 and IL-10R2. IL-22 signaling activates the JAK-STAT pathway and regulates genes involved in innate immunity, particularly in nonimmune cells such as keratinocytes and hepatocytes. It is co-expressed with IL-17A and IL-17F in Th17 cells and contributes to host defense, wound healing, and inflammatory responses.
