Target Information
MAP4K1 (also known as HPK1, Hematopoietic progenitor kinase 1) is a hematopoietic cell-restricted member of the Ste20 serine/threonine kinase superfamily. It functions as an upstream activator of the MEKK/JNK/SAPK signaling pathway and modulates T cell receptor (TCR) signaling and proliferation by phosphorylating the adaptor protein SLP-76.
JNKs/SAPKs play a key role in mammalian stress responses. MAP4K1 is predominantly expressed in hematopoietic progenitor cells and is distantly related to p21(Cdc42/Rac1)-activated kinase (PAK) and yeast STE20. HPK1 specifically activates JNK1 and enhances AP-1-mediated transcriptional activity. It binds and phosphorylates MEKK1 directly, and JNK1 activation by HPK1 is inhibited by dominant-negative MEKK1 or MKK4/SEK mutants. Unlike PAK65, HPK1 lacks the small GTPase Rac1/Cdc42-binding domain and does not bind Rac1 or Cdc42.
