Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. Upon stress, p53 activates transcription of genes such as p21waf1, leading to G1 or G2/M arrest.

p53 structure includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization, while Ser392 phosphorylation is associated with tumor formation. p53 is typically low in normal cells but highly expressed in transformed cells, contributing to malignancy. Mutations in TP53 can abolish DNA binding and tumor suppressor function, observed in many human cancers and genetic syndromes like Li-Fraumeni syndrome.
