Target Information
모든 생물은 고온 및 다양한 환경적 스트레스에 반응하여 열충격 RNA와 단백질을 빠르게 합성합니다. 열충격 유전자 전사는 열충격 반응 요소(HSE)에 결합하는 전사 활성화 인자인 열충격 인자(HSF)에 의해 조절됩니다. HSE는 5-뉴클레오타이드 {nGAAn} 모듈의 세 반복으로 구성되어 있으며, 모든 열충격 유전자 상류에 존재합니다. HSF는 종마다 분자량이 다르며, 면역학적 교차 반응이 거의 없습니다. 인간 세포에서는 HSF1과 HSF2가 확인되었으며, 두 인자는 동일한 HSE에 결합하고 38%의 서열 유사성을 가집니다. HSF1은 열, 중금속, 산화제 등 전형적인 스트레스 신호에 반응하며, 스트레스 시 핵 내 삼량체 형태로 활성화되어 DNA에 결합합니다. HSF1의 인산화는 열충격 유전자의 최대 전사 활성에 필수적입니다.
