• For IHC (PFA fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

Thyroid Peroxidase (TPO) encodes a membrane-bound glycoprotein enzyme essential for thyroid gland function. It catalyzes iodination of tyrosine residues in thyroglobulin and formation of phenoxy-ester bonds between iodinated tyrosines, producing thyroid hormones thyroxine (T4) and triiodothyronine (T3).
Mutations in TPO are associated with disorders such as congenital hypothyroidism, congenital goiter, and thyroid hormone organification defect IIA. Multiple transcript variants exist, though some full-length forms remain undetermined.
Diseases associated with TPO include Thyroid Dyshormonogenesis 2A and Congenital Hypothyroidism.
