Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.
Target Information
Thyroid Peroxidase (TPO) encodes a membrane-bound glycoprotein that functions as an enzyme essential for thyroid gland activity. It catalyzes iodination of tyrosine residues in thyroglobulin and formation of phenoxy-ester bonds between iodinated tyrosines, leading to thyroid hormone synthesis (thyroxine and triiodothyronine).
Mutations in TPO are associated with thyroid-related disorders such as congenital hypothyroidism, congenital goiter, and thyroid hormone organification defect IIA. Several transcript variants encoding distinct isoforms have been identified, though some full-length variants remain undetermined.
Diseases associated with TPO include Thyroid Dyshormonogenesis 2A and Congenital Hypothyroidism.
