Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. p53 up-regulates stress-responsive genes such as p21waf1, contributing to G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation sites on p53, including Ser15 and Ser392, are critical for activation, stabilization, and tumor formation processes. p53 functions as an anticancer agent by blocking proliferation of damaged cells and is implicated in aging, oxidative stress, and learning-related LTP deficits. It is expressed at low levels in normal cells but highly upregulated in transformed cell lines, contributing to malignancy. Mutations in TP53 lead to loss of tumor suppressor activity and are found in both somatic and germline forms, such as in Li-Fraumeni syndrome.
