Target Information

CtIP is a 125 kDa protein originally identified as interacting with the transcription repressor CtBP through the PLDLS motif (CtBP-Interacting Protein), suggesting a role in transcription regulation.
CtIP also interacts with BRCA1 through its C-terminal BRCT domains, indicating a potential tumor suppressor role.
This CtIP–BRCA1 interaction can be disrupted by DNA-damaging agents such as UV or gamma irradiation.
Li et al. (Nature 406, 210–215, 2000) demonstrated that ATM phosphorylates CtIP at serine residues 664 and 745. Mutation of these sites prevents BRCA1 dissociation from CtIP, leading to persistent repression of BRCA1-dependent induction of GADD45 upon ionizing radiation.
