Product Specific Information

Description:
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with CD3 gamma and delta, is essential for TCR assembly, trafficking, and surface expression. CD3 is expressed by thymocytes and all mature T cells. Binding of 145-2C11 to TCR can initiate intracellular signaling leading to activation, proliferation, or apoptosis depending on conditions. It is commonly used as a phenotypic marker for mouse T cells.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤1.0 µg per test (100 µL final volume). Cell number may vary from 10⁵ to 10⁸ per test. Optimize antibody titration for best performance.

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

Staining Recommendation:
Each NovaFluor conjugate or kit is shipped with CellBlox Blocking Buffer. Add 5 µL per sample when staining with NovaFluor conjugates or preparing antibody cocktails. For single-color controls, use 5 µL CellBlox per 100 µL of cell sample (10³–10⁸ cells).

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into the cytoplasm of T cells and regulating TCR surface expression. CD3 consists of gamma, delta, epsilon, and zeta subunits, which are structurally related members of the immunoglobulin superfamily encoded on chromosome 11. These subunits associate with cytoplasmic signaling molecules via tyrosine-based motifs. CD3 plays a role in TCR-induced growth arrest, survival, and proliferation. It is expressed on 68–82% of normal peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Defects in CD3 genes are associated with CD3 immunodeficiency.
