Target Information

IL-33R, also known as ST2, is a receptor for IL-33 and a member of the IL-1 receptor family. Its extracellular domain has three Ig-like domains that interact with its co-receptor IL-1RAcP and the ligand IL-33. Upon IL-33 binding, the IL-33R complex associates with adaptor proteins MyD88, IRAK1, IRAK4, and TRAF6, activating NF-κB and MAPK pathways. This leads to the release of cytokines such as IL-5, IL-6, IL-8, and IL-13. Two main isoforms exist: ST2L (membrane-bound) and ST2S (soluble). The soluble form acts as a decoy to dampen IL-33 signaling. IL-33R is expressed mainly by mast cells, basophils, and eosinophils, and in mice, on ILC2 and Th2 cells. IL-33R signaling is implicated in immune conditions such as allergies, arthritis, atherosclerosis, and sepsis.
