Target Information
COX2 converts arachidonate to prostaglandin H2 (PGH2), a key step in prostanoid synthesis and inflammatory prostaglandin production. The reaction proceeds via a cyclooxygenase (COX) activity forming PGG2, followed by a peroxidase activity reducing PGG2 to PGH2.
COX2 is constitutively expressed in certain tissues (endothelium, kidney, brain) and up-regulated under pathological conditions such as cancer and inflammation. Increased COX2 levels are linked to enhanced cell adhesion, resistance to apoptosis, and tumor angiogenesis.
COX2 is inhibited by calcitriol (active Vitamin D) and glucocorticoids can repress PTGS2 gene activity. NSAIDs such as aspirin target COX2 to reduce pain and inflammation.
