Target Information

Keyhole limpet hemocyanin (KLH) is a large, heterogeneous glycosylated protein composed of subunits with molecular weights of 350,000 and 390,000, forming aggregates of 4,500,000–13,000,000 Da. Each domain contains two copper atoms that bind one oxygen molecule (O₂), giving the molecule a transparent blue color when oxygenated.

KLH is highly immunogenic yet safe in humans, making it a preferred carrier protein in vaccine and antibody production. It cannot be synthesized artificially and is purified from Megathura crenulata (keyhole limpet). KLH is widely used as a carrier for haptens—small molecules that require conjugation to a larger protein to elicit an immune response.

Its large size and multiple epitopes generate a strong immune response, while abundant lysine residues facilitate high hapten-to-carrier ratios. Because KLH is derived from a gastropod, it is phylogenetically distant from mammalian proteins, reducing cross-reactivity in mammalian systems. However, its tendency to aggregate can complicate laboratory handling, though aggregates remain immunogenic.
