Store vial at 4°C prior to restoration. For extended storage, aliquot contents and freeze at -20°C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use.

Anti-Thymidylate Synthase IgG fraction is directed against human Thymidylate Synthase and is useful in detecting its presence in various assays. Because inhibition of Thymidylate Synthase prevents DNA synthesis and cell proliferation, the enzyme is an important target for cancer chemotherapeutic drugs, especially fluoropyrimidine antineoplastic drugs used to treat solid tumors. This antibody can detect antigen in a variety of human cells and tissues, as well as bacteria, African green monkey, rat, and mouse. Lower dilutions may be required in non-human cell lines. Anti-Thymidylate Synthase detects Thymidylate Synthase by immunochemistry in proliferating cell cultures and tissues but does not stain non-proliferating cells. Normal colon mucosa shows weak staining, while some colorectal cancer specimens show very strong staining.

Target Information

Thymidylate Synthase (TS) converts deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP), essential for DNA biosynthesis. TS is a critical target for fluoropyrimidines, an important group of antineoplastic drugs used in solid tumor treatment. Both 5-FU and fluorodeoxyuridine are converted in tumor cells to FdUMP, which inactivates TS by forming a ternary covalent complex with the folate cofactor 5,10-methylenetetrahydrofolate. Expression of TS protein is associated with response to 5-fluorouracil (5-FU) in human colorectal, gastric, head and neck, and breast carcinomas.
