Target Information
TPA converts the abundant but inactive zymogen plasminogen to plasmin by hydrolyzing a single Arg-Val bond in plasminogen. By controlling plasmin-mediated proteolysis, it plays an important role in tissue remodeling, degradation, cell migration, and various physiological and pathological events.
It specifically cleaves the Arg-Val bond in plasminogen to form plasmin and is composed of a heterodimer of chain A and chain B connected by a disulfide bond.
TPA binds to fibrin with high affinity, which increases the catalytic efficiency between 100–1000-fold by enhancing affinity for plasminogen. Binding to heparin also increases plasminogen activation.
Additionally, TPA binds to laminin, fibronectin, mannose receptor, and LRP1, which are involved in TPA clearance. It also interacts with annexin II and cytokeratin 8.
Unidentified interactions on endothelial and vascular smooth muscle cells lead to a 100-fold stimulation of plasminogen activation, while binding to VSMC reduces TPA inhibition by PAI-1 approximately 30-fold.
