Target Information
STAT4 was originally identified using degenerate primers complementary to conserved regions of other STAT proteins. STAT4 shares similarity with STAT1 (52%) and STAT3 (47%), and can be tyrosine phosphorylated by Jak1 or Jak2. Activated STAT4 forms homo- or heterodimers and binds the IFN-gamma activated site (GAS). Serine phosphorylation is also required for maximal transcriptional activity.
STAT4 expression is mainly found in thymus, spleen, and testis. It is activated by interleukin 12 (IL-12), which is essential for T-cell independent induction of IFN-gamma and Th1 differentiation. STAT4-deficient mice show impaired Th1 development and enhanced Th2 response. In viral infection, IFN-α/β activation of STAT4 is required for IFN-γ production.
