For IHC(P), use heat-induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen.
Target Information
Smoothened drosophila homolog (SMOH) is a serpentine protein belonging to the G-protein coupled receptor Fz/Smo family with 787 amino acids. It contains seven transmembrane domains, four glycosylation sites, and a putative extracellular amino terminus.
Smoothened is activated by β-Arrestin 2 and GRK2, acting as a receptor for Sonic hedgehog (Shh), and serves as a positive regulator of the hedgehog signaling pathway. Together with patched protein (PTCH), it regulates vertebrate developmental processes, cellular proliferation, and differentiation by activating transcription factor Gli.
Mutations in Smoothened are associated with sporadic basal cell carcinoma of the skin. It is widely expressed in heart and brain tissues.
