Target Information

Oncogene ERBB4 (HER4) is a member of the type I receptor protein tyrosine kinase subfamily, which includes EGFR, ERBB2, and ERBB3. ERBB4 binds and is activated by neuregulins and other factors, inducing various cellular responses such as mitogenesis and differentiation.
ErbB4 acts as a receptor for heregulin and mediates HGL-stimulated tyrosine phosphorylation. It plays a key role in signal transduction related to metabolism, transcription, cell cycle progression, cytoskeletal rearrangement, movement, apoptosis, and differentiation.
Expression of ErbB4 is high in several breast carcinoma cell lines (T47-D, MDA-MB-453, BT-474, H3396) and in normal tissues such as skeletal muscle, heart, pituitary, brain, and cerebellum. Intermediate expression is found in MCF-7, MDA-MB-330, and MDA-MB-361, while low or absent expression is observed in MDA-MB-231, MDA-MB-157, MDA-MB-468, and SKBR-3. Elevated ErbB4 levels in some tumor cell lines suggest a potential role in malignancy.
