LRP6는 Wnt/β-catenin 신호전달 경로에 필수적인 단백질로, Frizzled와 함께 Wnt의 코리셉터로 작용합니다. DKK1 및 DKK2에 대한 고친화성 수용체이며, DKK1과의 상호작용은 Kremen 단백질 매개 엔도사이토시스를 통해 LRP6 매개 신호를 억제합니다.
