SMC6은 SMC5-SMC6 복합체의 핵심 구성 요소로, 상동 재조합을 통한 DNA 이중가닥 절단 복구에 관여합니다. 이 복합체는 SMC1-SMC3 cohesin 복합체를 손상 부위로 유도하여 자매 염색분체 간 상동 재조합을 촉진할 수 있습니다.
또한 ALT(Alternative Lengthening of Telomeres) 세포주에서 재조합을 통한 텔로미어 유지에 필요하며, shelterin 복합체 성분의 sumoylation을 매개하여 ALT 관련 PML body(APB)에서 shelterin 복합체 해체를 유도하는 것으로 제안됩니다.
SMC5-SMC6 복합체는 원형 바이러스 DNA와 같은 episomal DNA의 전사를 억제할 수도 있습니다.
