Product Specific Information

The LG.7F9 monoclonal antibody reacts with mouse CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by nearly all mature T cells and a subpopulation of B cells, mainly memory B cells. In mouse, CD27 is found on most thymocytes except for CD46-CD8- precursors. CD27 interacts with CD70, playing a critical role in T cell–B cell interactions and effector cell maturation. LG.7F9 cross-reacts with human and rat CD27.

Applications Tested: Tested on mouse splenocytes; ≤0.5 µg per test (100 µL volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.

Fluorochrome Information:
PE-eFluor 610 is excited by 488–561 nm lasers and emits at 607 nm. Use a 610/20 band pass filter (equivalent to PE-Texas Red). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples may be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

CD27 is a 50 kDa member of the TNF receptor superfamily, associated with CD40 and CD30. It regulates cell proliferation and death, and its expression is confined mainly to lymphocytes. CD27 is expressed as a disulfide-linked homodimer on mature thymocytes, T cells, and a subset of B cells. Upon TCR-CD3 activation, CD27 expression increases, and soluble CD27 (sCD27) is released, serving as a prognostic marker in B cell malignancies. CD27-binding protein (SIVA) mediates apoptosis through CD27 signaling. Dysregulation of CD27 is linked to Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
