Product Specific Information

Description:
The HIT2 monoclonal antibody reacts with the human CD38 molecule, an approximately 45 kDa type II transmembrane protein expressed by thymocytes, peripheral B cells (including plasma cells), activated T cells, and monocytes. CD38 is a counter-receptor for CD31 and exhibits cyclase and hydrolase enzymatic activity.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorophore Information:
PE-eFluor® 610 is excited by 488–561 nm laser lines and emits at 607 nm. Recommended detection filter: 610/20 band pass (equivalent to PE-Texas Red®).
Light sensitivity: Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Excitation/Emission:
488–561 nm / 607 nm (Blue, Green, Yellow-Green Laser)

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD38 (NAD⁺ glycohydrolase):
A type II transmembrane glycoprotein involved in activation, proliferation, and differentiation of lymphocytes, and apoptosis of progenitor cells. Functions as a multi-catalytic ectoenzyme (ADP-ribosyl cyclase, cyclic ADP-ribose hydrolase, NAD⁺ glycohydrolase) and as a cell surface receptor.
Antibodies to CD38 are used for:

• Subtyping of lymphomas and leukemias
• Detection of plasma cells (e.g., myelomas)
• Marker for activated B and T cells

CD38 participates in cell adhesion, signal transduction, and calcium signaling. Highly expressed in pancreas, liver, kidney, malignant lymphoma, and neuroblastoma.
Associated diseases include chronic lymphocytic leukemia and Richter’s Syndrome.
