Target Information

KLRB1 (Killer cell lectin-like receptor subfamily B, member 1, CD161) is a type II membrane protein with an external C terminus. It is expressed on nearly all NK cells and a subset of CD3⁺ T cells.
KLRB1 forms a homodimeric cell surface protein (40–44 kDa) and plays an inhibitory role in NK cell activity. Activation of KLRB1 stimulates acid sphingomyelinase/SMPD1 and increases intracellular ceramide, leading to AKT1/PKB and RPS6KA1/RSK1 kinase activation and T cell proliferation.
KLRB1 acts as a lectin binding to Gal-α(1,3)Gal and N-acetyllactosamine epitopes, as well as CLEC2D/LLT1 ligand, thereby inhibiting NK cell-mediated cytotoxicity and interferon-γ secretion.
The protein includes an extracellular domain with C-type lectin motifs, a transmembrane domain, and a cytoplasmic domain.
