Target Information
STK3 and STK4 are stress-activated, pro-apoptotic kinases that, following caspase cleavage, translocate to the nucleus to induce chromatin condensation and DNA fragmentation. They are key components of the Hippo signaling pathway, which regulates organ size and tumor suppression by controlling proliferation and apoptosis.
The core Hippo pathway involves a kinase cascade where STK3/MST2 and STK4/MST1, with regulatory protein SAV1, activate LATS1/2 and MOB1. This leads to phosphorylation and inactivation of the YAP1 oncoprotein, preventing its nuclear translocation and regulating genes involved in proliferation, cell death, and migration.
STK3/MST2 and STK4/MST1 repress mature hepatocyte proliferation, prevent activation of oval cells, and inhibit tumor formation. They phosphorylate NKX2-1, NEK2, MOBKL1A, RASSF2, and MOBKL1B, and regulate centrosome disjunction and ESR1 transcriptional activity. They also positively regulate RAF1 activation by suppressing inhibitory phosphorylation on Ser-259.
