Target Information
STK3 and STK4 are stress-activated, pro-apoptotic kinases. Upon caspase cleavage, they translocate to the nucleus and induce chromatin condensation and DNA fragmentation.
They are key components of the Hippo signaling pathway, which regulates organ size and tumor suppression by controlling proliferation and apoptosis.
STK3/MST2 and STK4/MST1 form complexes with SAV1 to phosphorylate and activate LATS1/2–MOB1, which in turn inactivates YAP1 and WWTR1/TAZ.
These kinases are essential for:
- Repressing hepatocyte proliferation
- Preventing activation of adult liver stem cells
- Inhibiting tumor formation
- Phosphorylating NKX2-1, NEK2, MOBKL1A/B, RASSF2
- Regulating centrosome disjunction and RAF1 activation
