Product Specific Information

Description:
The IP26 monoclonal antibody reacts with the human alpha beta TCR dimer, expressed by the majority of peripheral T cells.

Applications Reported:
This antibody is reported for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.5 µg) per test (defined as staining a cell sample in 100 µL final volume).
Cell numbers can range from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm.
Recommended filter: 610/20 bandpass.
Ensure instrument compatibility before use.

Staining Recommendation:
When using multiple Super Bright conjugates, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product # 00-8222)
• 1-step Fix/Lyse Solution (Product # 00-5333)

Target Information

The T cell antigen receptor (TCR) is a ligand-specific alpha/beta heterodimer associated with invariant CD3 chains (gamma, delta, eta, zeta).
These components are essential for surface expression and signal transduction during T cell activation.
Most T cells in blood and lymphoid tissues express TCR α/β, while γ/δ T cells are mainly epithelial.
TCR engagement leads to differentiation, proliferation, and cytokine production.
Defects or autoantibodies against TCR components are associated with autoimmune diseases such as RA and SLE.
TCR recognition of self-peptides is linked to autoimmunity, while mutant self-peptides are implicated in tumor immunity.
