Product Specific Information
- IHC (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- Formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD31 (platelet endothelial cell adhesion molecule-1, PECAM-1) is an inhibitory coreceptor that regulates T and B cell signaling through dual immunoreceptor tyrosine-based inhibitory motifs (ITIMs). Upon phosphorylation by associated kinases, these motifs provide docking sites for protein-tyrosine phosphatases.
CD31 is ubiquitously expressed within the vascular compartment, primarily at junctions between adjacent cells. Its N-terminal Ig-like domain mediates homophilic binding, essential for cell–cell interactions.
Functions of CD31 include:
- Modulation of integrin-mediated cell adhesion
- Regulation of transendothelial migration and angiogenesis
- Involvement in apoptosis and immune regulation
- Roles in autoimmunity, macrophage phagocytosis, IgE-mediated anaphylaxis, and thrombosis
