Product Specific Information

The PA5-20007 immunogen is located within the last 50 amino acids of PUMA. The immunogenic sequence is identical between the A and B forms of PUMA.
In Western blot applications, this antibody detects a band at approximately 23 kDa. A suggested positive control is K562 cell lysate.
PA5-20007 can be used with blocking peptide PEP-0127.
Predicted species reactivity based on immunogen sequence: Rat (100%), Mouse (100%).


Target Information

PUMA is a pro-apoptotic Bcl-2 family member, along with Bax and Noxa, which are transcriptional targets of p53. The PUMA gene encodes two BH3 domain-containing proteins, PUMA-a and PUMA-b. These proteins bind Bcl-2, localize to mitochondria, and induce cytochrome c release and apoptosis in response to p53.
PUMA may act as a direct mediator of p53-induced apoptosis. Antisense inhibition of PUMA expression reduces the apoptotic response to p53, suggesting PUMA’s role in mediating p53-dependent cell death via the cytochrome c/Apaf-1 pathway.
