Target Information

Histone H2A.X (H2AX) is a member of the histone H2A family, one of the four core histones forming the nucleosome core particle. DNA double-strand breaks (DSBs) trigger phosphorylation of serine 139 at the carboxy terminus of H2AX, producing gamma-H2AX, a marker of DSBs detectable by Western blotting or immunofluorescence. Phosphorylation is mediated by phosphatidylinositol 3-kinases, with ATM, DNA-PK, and ATR implicated in this process. H2A.X consists of 143 amino acids and is synthesized during G1 and S phases. Its mRNA exists in two forms—one polyadenylated (~1600 bases) and one non-polyadenylated (~575 bases). The gene maps to human chromosome 11q23.2–q23.3 and plays a role in nucleosome formation and DNA structure. The H2A.X variant regulates the interaction of MDC1, a DNA repair protein, at damage sites.
