Alzheimer’s disease (AD) 환자 중 일부는 presenilin 단백질(PSEN1 또는 PSEN2) 또는 amyloid precursor protein (APP)의 유전적 변이를 가지고 있습니다. 이러한 변이는 amyloid-beta의 긴 형태 생산을 증가시켜, AD 뇌에서 발견되는 amyloid 침착의 주요 성분이 됩니다. Presenilin은 gamma-secretase 효소의 활성 조절을 통해 APP 처리에 영향을 미치며, Notch 수용체 절단에도 관여하는 것으로 알려져 있습니다. PSEN2는 두 가지 대체 스플라이스 변이를 가지며, 서로 다른 isoform을 생성합니다.
