Product Specific Information
- For immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
- For formalin-fixed tissues: heat tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, then cool at room temperature for 20 min.
Target Information
BCL-2 is a key regulator of apoptosis that functions to inhibit or promote cell death. Members of the BCL-2 family can dimerize to modulate apoptosis. For example, Bag1 forms a heterodimer with BCL-2 to enhance its anti-apoptotic effect. Bax and Bak are critical in cytochrome c release from mitochondria, initiating apoptosis. Constitutive expression of BCL-2, such as by translocation to the Ig heavy chain locus, is associated with follicular lymphoma. Neoplastic germinal centers in such lymphomas express high levels of BCL-2 alpha protein, unlike normal or hyperplastic centers. Two transcript variants of BCL-2 differ at their C-terminal ends. Overexpression of BCL-2 is linked to cancers such as B-cell lymphoma and prostate cancer.
