Target Information

HMGB1 (High-mobility group box-1) protein was originally described as a nuclear non-histone DNA-binding chromosomal protein. Recent studies show that damaged or necrotic cells release HMGB1 extracellularly, where it functions as a proinflammatory cytokine.
Mouse HMGB1 is a 215 amino acid single-chain polypeptide with three domains: two positively charged DNA-binding domains (HMG box A, aa 9–79; box B, aa 89–162) and a negatively charged 30 aa C-terminal acidic tail. Residues 28–44 and 180–185 contain nuclear localization signals (NLS).
The cytokine activity resides in the B box, while the A box interacts with transcription factor domains. HMGB1 mediates inflammation, monocyte migration, cytokine secretion, and T cell–dendritic cell interactions. It signals through receptors such as RAGE, TLR2, and TLR4.
HMGB1 is highly conserved, present in nuclei and cytoplasm of most cell types, and is critical in sterile and infection-associated inflammatory responses. It also acts as a DNA-binding protein and an early trigger of sterile inflammation via TLR4 and RAGE activation. Elevated HMGB1 levels are associated with tissue injury, infection, endotoxemia, sepsis, arthritis, and ischemia-reperfusion injury.
