Target Information
The HSP70 family comprises highly conserved proteins induced by various biological stresses, including heat stress, across all examined organisms. Human HSP70 family members include:
- HSP70: Strongly inducible in all organisms, constitutively expressed in primate cells
- HSP72: 72 kDa protein induced exclusively under stress conditions
- HSC70 (cognate protein): 72 kDa constitutively expressed protein involved in uncoating of clathrin-coated vesicles
- GRP78 (BiP): Glucose-regulated 78 kDa protein localized in the endoplasmic reticulum
- p75 (HSP75): 75 kDa protein found within mitochondria
HSC70 (also known as HSC71, HSC73, HSP73, p72, prp73) is constitutively expressed and slightly heat-inducible. It binds to exposed loops of clathrin light chains to promote uncoating and can also interact with the cytoskeleton, facilitating cytoskeletal rearrangements. HSC70 stimulates lysosomal degradation of intracellular proteins and retards aggregation and folding of mitochondrial precursor proteins in vitro.
