Target Information
The Histamine H4 receptor has been reported in human peripheral blood leukocytes, bone marrow, colon, liver, lung, small intestine, spleen, testis, thymus, tonsil, and trachea. Unlike the mouse H4 receptor, the human gene was not detected in brain.
G-protein Coupled Receptors (GPCRs) comprise one of the largest families of signaling molecules, with over a thousand members predicted to exist. All GPCRs share a structural motif of seven membrane-spanning helices and exist in both active and inactive forms. Ligand binding induces conformational changes leading to signaling via G-proteins and downstream effectors.
Studies on negative regulation of GPCRs reveal mechanisms critical to cellular physiology and pharmacological research. Notable GPCR-targeted drugs include Zyprexa (Eli Lilly) and Claritin (Schering-Plough), widely used in mental health and allergy treatments.
