Target Information
In the intact cell, DNA closely associates with histones and other nuclear proteins to form chromatin. Chromatin remodeling is critical for transcriptional regulation, and histone acetylation plays a major role in this process. Acetylation of lysine residues in histone tails induces conformational changes, increasing DNA accessibility to transcription factors.
Histone acetylases include GCN5, PCAF, p300/CBP, HAT1, and TAF II p250. Conversely, histone deacetylases (HDACs) such as HDAC1–9 mediate transcriptional silencing. HDAC9 and HDAC9a are alternatively spliced isoforms; HDAC9a is 132 amino acids shorter but retains catalytic activity. Both isoforms repress myocyte enhancer-binding factor 2-mediated transcription and are expressed in brain, skeletal muscle, kidney, placenta, and pancreas.
