Product Specific Information

Description:
The PC61.5 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early progenitors of T and B lineage as well as by activated mature T and B lymphocytes. CD25 binds IL-2 with low affinity alone, but forms a high-affinity receptor when associated with CD122 (IL-2Rβ) and CD132 (common γ chain). Binding of IL-2 to both high and low affinity receptors is inhibited by the PC61.5 antibody. CD25 is involved in lymphocyte differentiation and activation/proliferation.

Applications Reported: Flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.125 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize by titration for best performance.

Fluorophore Information:
Super Bright 436 is excited by the violet laser (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

CD25 (IL2RA):
Cytokine receptor subunit involved in T and B lymphocyte proliferation. The high-affinity IL-2 receptor is composed of IL2RA, IL2RB, and IL2RG chains. IL2RA homodimers form low-affinity receptors; IL2RB homodimers form medium-affinity receptors. IL2 plays key roles in growth, differentiation, and apoptosis regulation of immune cells. Mutations in IL2RA are associated with immune dysregulation disorders such as interleukin 2 receptor alpha deficiency.
