Target Information

ACAP2 is a transport carrier belonging to the Centaurin family with three ANK repeats, an Arf-GAP domain, a BAR domain, and a PH domain.
It functions as a GTPase-activating protein (GAP) for ADP-ribosylation factor 6 (ARF6) with robust constitutive ARF6 GAP activity.
This activity is stimulated by phosphatidylinositol 4,5-bisphosphate (PIP2) and phosphatidic acid, but has little activity toward ARF1.
ACAP2 controls the return of ARF to the inactive GDP-bound state and, along with ACAP1, is targeted by phosphatidylinositol (3,4,3) triphosphate to inhibit PDGF-induced dorsal membrane ruffles in NIH3T3 fibroblasts when overexpressed.
ACAP2 is phosphorylated upon DNA damage, likely by ATM or ATR, and shows widespread expression, with the highest level in lung tissue.
